چکیده

Abstract INTRODUCTION Comprising approximately 8% of our genome, Human Endogenous RetroViruses (HERVs) represent a class germline retroviral infections that are regulated through epigenetic modifications. In cancer cells, which often have dysregulation, HERVs been implicated as potential oncogenic drivers. However, their role in gliomas is not known.Given the link between HERV expression cell lines and distinct dysregulation gliomas, we utilized tailored bioinformatic pipeline to characterize validate glioma retrotranscriptome correlate with locus-specific modifications.Method:A custom workflow was used quantify interest. Cell-line methylation quantified using script. We generated primers specific for endogenous Mouse mammary tumor (MMTV)-Like virus 6 (HML-6). Visualization RNA transcripts performed RNA-scope. Clinical data obtained R package, TCGABiolinks. RESULTS The A172 line had significantly higher mean overall relative M059J H4 (p< 0.0001 both). cells lower number CpG islands There significant inverse correlation beta value FC (R=-0.57, p=0.01). qPCR confirmed robust HML-6 locus culture neurospheres. Elevated ERVK3-1 associated reduced survival among IDHwt GBM patients (18.3 vs. 15.1 months, p=0.039). This preserved IDH mutant (IDHm) well (17.9 months 14.0 p=0.0088). CONCLUSION correlates loss DNA at loci. overexpressed highly invasive glioblastoma patient-derived demonstrated detriment elevated product, ERVK3-1.

Prognosis and Survival Study in Patients with Glioblastoma Multiform and Its Relationship with EGFR Expression

Background and Aim: Glioblastoma multiforme (GBM) is the most common malignant and invasive tumor of the brain. The relation between prognosis and survival of GBM patients with Epidermal Growth Factor Receptor (EGFR) expression is challenging. Thus, we aimed to evaluate the prognosis and survival of patients with GBM and its relationship with EGFR expression. Materials and Methods: This single...

Expression of HERV-Fc1, a human endogenous retrovirus, is increased in patients with active multiple sclerosis.

Multiple sclerosis (MS) is considered to be an autoimmune disease with an unknown cause and with immune system dysregulation. Among environmental factors, viruses are most often connected with the etiology of MS. Human endogenous retroviruses (HERVs) constitute 5 to 8% of human genomic DNA and have been detected as transcripts and proteins in the central nervous system (CNS) and peripheral bloo...

Elevated HERV-K Expression in Soft Tissue Sarcoma Is Associated with Worsened Relapse-Free Survival

A wide variety of endogenous retroviral sequences has been demonstrated in the human genome so far, divided into several different families according to the sequence homology to viral strains. While increased expression of human endogenous retrovirus (HERV) elements has already been linked to unfavorable prognosis in hepatocellular carcinoma, breast cancer, and ovarian carcinoma yet less is kno...

Endogenous antibody responsiveness to epidermal growth factor receptor is associated with immunoglobulin allotypes and overall survival of patients with glioblastoma.

BACKGROUND Immunoglobulin γ marker (GM) and κ marker (KM) allotypes, hereditary antigenic determinants of γ and κ chains, respectively, have been shown to be associated with immunity to a variety of self and nonself antigens, but their possible contribution to immunity to the tumor-associated antigens epidermal growth factor receptor (EGFR) and EGFR variant (v)III has not been evaluated. The ai...
